Modified reagents for determination of urea and ammonia.

چکیده
This reduces the number of reagents required for color development to two, each of which is stable for 60 days or more if kept cool and in amber bottles protected from light. These reagents are added in equal volumes to the ammonia sample. For example, to 1-5 g. of ammonia in a volume of 1 ml. or less, 5 ml. each of diluted solutions 1 and 2 are added successively. Maximum blue color is produced at room temperature in about 30 mm. and the absorbance remains unchanged for 24 hours or more. Alternatively, one may use 1 ml. each of the more concentrated reagents and develop the color at a higher temperature-for example, at 50-60#{176}. In this case, color development will be completed in about 3

Quantitative determination of urea concentrations in cell culture medium.
Urea is the major nitrogenous end product of protein metabolism in mammals. Here, we describe a quantitative, sensitive method for urea determination using a modified Jung reagent. This assay is specific for urea and is unaffected by ammonia, a common interferent in tissue and cell cultures. We demonstrate that this convenient colorimetric microplate-based, room temperature assay can be applied...
متن کاملDirect fluorometric determination of urea in urine.
Urea in biological fluids has been determined by a variety of direct and indirect procedures. Some indirect methods depend on the hydrolysis of urea with the enzyme urease to form ammonia, with subsequent quantitation of the ammonia by nesslerization (1-4). Undesirable features of these indirect methods are the use of relatively unstable reagents and loss of ammonia during the procedure. Direct...
متن کاملDetermination of Ammonia in Blood by Donald D. Van Slyke and Alma Hiller with the Technical Amstance of John Plazin
In the method here presented Nash and Benedict’s (1921) procedure for aerating the ammonia from blood into an acid-receiving solution is utilized without modification. For determining the ammonia obtained, however, we have employed, instead of Nesslerization, the blue color developed when ammonia is heated with an alkaline solution of phenol and hypochlorite. This reaction was apparently first ...

METHOD for the miieasureniemit of seiummmadenosiime deaminase activity was recently published in this ,Journal (1). This method requires 2 ml. of serum, suffers froni ammunnecessarily large number of manipulations, ahl(l utilizes aim unstable, relatively insensitive Nessler color reaction (2). Attempts to overcome these difficulties constitute the basis for this report. A catalyzed indopheimol ...
